Inter-subunit contacts form the allosteric activator binding site and are not conserved in BsuNagB. In particular, residues of 7 and its preceding loop eg. Ser151, Arg158 and Lys160 ; , and the C-terminal -helix extension eg. Tyr254 ; are characteristic of this hexameric group. The 6-PGL structure rmsd between Tma6PGL and BsuNagB is 1.67 over 185 equivalent C atoms, Figure 3 ; also allows us to dissect out the conserved structural and catalytic features. Across the 3 groups, equivalent catalytic side chains are conserved, ie. Fortunately, even before I had succeeded in convincing him of the undesirability of our getting involved in Spiritualism, there had occurred the first of a whole chain of events that was to be of far greater value to us, spiritually speaking, than any number of sessions with the Ouija board. Indeed, they were to constitute the most interesting and worthwhile part of our stay in Bangalore, and an important stage of our pilgrimage. On our first or second Sunday in Rajgopals house, in the morning, four or five young men came to see us. In this there was nothing unusual. What was unusual, however, was the way in which they paid their respects to us. Instead of the usual salutation with folded hands, or more or less perfunctory touching of the feet, each of them in turn went through an elaborate full-length prostration that lasted several minutes and was quite different from anything we had ever seen before. Arms were raised above the head and lowered again, body was spread-eagled on the floor, head turned from side to side, arms extended and withdrawn one after the other in wide sweeping movements, the whole exercise being performed slowly and with great concentration. More remarkable still, all the young men wore Western dress and all were educated, that is to say, could read, write, and speak English. When the prostrations had been completed we entered into conversation with them, making the standard Indian enquiries, and it transpired that they were disciples of an ascetic popularly known as Yalahankar Swami. Yalahankar, they told us, was the name of a small town some thirty miles from Bangalore, where the swami had his ashram, and where he usually stayed. On hearing that we were staying with Rajgopal he had told them to go and pay their respects to us. The young men themselves lived in Bangalore, where, it seemed, they all had good jobs. When we asked them the meaning of their unusually elaborate prostrations they explained that according to Yalahankar Swami the majority of people, even religious people, were full of egotism. On account of this egotism they generally paid their respects to ascetics and holy men in a casual, hasty, and perfunctory manner. Since the purpose of the custom was to help curb egotism, to pay ones respects in this sort of way was of no value whatsoever. Indeed, by making people think how religious they were, or how humble, it could even intensify egotism. The swami had therefore devised for the use of his disciples the long and elaborate type of prostration we had just witnessed, and insisted that whenever they met a sadhu they should pay their respects to him by making it a number of times. The infection of E. coli with the DNA containing bacteriophage T2 was shown, by the classical experiments of Volkin and Astrachan, 10 to result in the formation of an unstable RNA fraction with a base composition which reflects that of the phage DNA. It is now generally accepted that this is the mRNA which carries the genetic information for the production of phage-specific proteins. The effect of levallorphanloa on the production of phages X and T6 in E. coli K-13 was tested. In the presence of 1.3 X 10-3 M levallorphan the yield of T6 particles per infected cell was 100, as compared to 130 in the absence of the drug. The corresponding values for the yield of X were 11 and 14, respectively. Thus, levallorphan, at a concentration at which we found it to inhibit RNA synthesis profoundly, had no significant effect on the production of DNA bacteriophages and therefore on the synthesis of their messenger RNA. Evidence for the synthesis of transfer RNA sRNA ; in the presence of levorphanol: Figure 2 shows the results of an experiment in which E. coli K-13 was incubated with C14-uracil in the presence and absence of levorphanol for 60 min followed by a 15-mim exposure to a large excess of C12-uracil. This chasing procedure results in the labeling of ribosomal and transfer RNA with little radioactivity in the rapidly turningover messenger fraction. RNA isolated from these cultures by the phenol procedure was fractionated in a sucrose gradient. It can be seen that C14-uracil incorporation into the 23S and 16S ribosomal RNA peaks is inhibited to a much greater extent than incorporation into the 4S transfer RNA peak.
